Pharmacology

This vaccine is used for the active immunisation of diphtheria, tetanus, pertussis, and poliomyelitis.

Mechanism of action

The vaccine stimulates the production of specific antibodies against diphtheria, tetanus, pertussis, and poliomyelitis by introducing antigens and triggering an immune response.

Dosage

Child (Primary vaccination): 3 injections (0.5 mL) at 2, 3, 4 months (1-month intervals).
Booster vaccination: 1 injection one year after primary vaccination (usually 16-18 months); 1 injection between 5-13 years.

Administration

Administer as a deep intramuscular injection (antero-lateral thigh in infants, deltoid area in children).

Missed dose

The vaccine is administered by healthcare professionals. You cannot miss a dose yourself.

Side effects

Common: Local injection-site reactions (pain, redness, swelling), abnormal crying, loss of appetite, irritability (usually within 48 hours, may continue for 48-72 hours).
Booster vaccination: Frequency of injection-site reactions tends to increase.
Children aged 2 years or more: Myalgia, malaise, headache.

Precautions & warnings

Immunogenicity may be reduced by immunosuppressive treatment or immunodeficiency; wait until end of treatment. Vaccination recommended in HIV infection. Caution in Guillain-Barre syndrome or brachial neuritis history. Caution in thrombocytopenia or bleeding disorders.

Contraindications

Hypersensitivity (active substances, excipients, glutaraldehyde, neomycin, streptomycin, polymyxin B, pertussis vaccine); life-threatening reaction after previous vaccination; febrile or acute disease; evolving encephalopathy; encephalopathy within 7 days of previous pertussis-containing vaccine.

Drug interactions

M-M-RVAXPRO or HBVAXPRO: Can be administered simultaneously but at separate sites.
Haemophilus influenzae type b conjugate vaccine (Act-HIB): Can be associated or combined.

Food interactions

Can be used with or without food.
