Eltrombopag Olamine is a thrombopoietin agonist used for the treatment of thrombocytopenia in chronic immune thrombocytopenic purpura (ITP), chronic hepatitis C infection, and severe aplastic anemia.

- Thrombocytopenia in persistent or chronic immune thrombocytopenia

Pharmacology
Eltrombopag Olamine is a small molecule thrombopoietin (TPO) receptor agonist. It interacts with the transmembrane domain of the human TPO-receptor and initiates signaling cascades that induce proliferation and differentiation from bone marrow progenitor cells, finally leading to increased platelet production.
Dosage
Adult Dose:
- Chronic ITP: 50 mg once daily, adjust to maintain platelet count ≥50×109/L. Max: 75 mg/day.
- Chronic hepatitis C: 50 mg once daily, max 100 mg/day.
- Severe aplastic anemia: 50 mg once daily, adjust to maintain platelet count ≥50×109/L. Max: 150 mg/day.
Child Dose (6 years+): 50 mg once daily. (1-5 years: 25 mg once daily).
Side effects

Use in pregnancy
Eltrombopag Olamine may be unsafe during pregnancy. Animal studies have shown harmful effects. Doctor will weigh benefits and risks.
Pregnancy Category: C — always consult a doctor before taking during pregnancy.
Precautions & warnings
- Eltrombopag Olamine can cause dizziness and make you less alert, affecting driving ability.
- Take on empty stomach (1 hour before or 2 hours after meals).
- Take at least 2 hours before or 4 hours after dairy products and calcium-fortified juices.
- Monitor platelet counts regularly.
- Monitor liver function.
Overdose effects
Overdose may cause excessive platelet count increase leading to thrombotic or thromboembolic complications. Consider oral administration of metal cation-containing preparation (calcium, aluminium or magnesium) to chelate eltrombopag and limit absorption.
