Description

Adapalene 0.1% + Benzoyl Peroxide 2.5% is a combination gel used for the treatment of acne vulgaris. Adapalene is a retinoid that binds to specific retinoic acid nuclear receptors, normalizing follicular epithelial cell differentiation and reducing microcomedone formation. Benzoyl Peroxide is an oxidizing agent with bacteriocidal and keratolytic effects. It removes dead skin cells and kills acne-causing bacteria. Apply once daily at night.

Acne vulgaris, in patients 12 years of age and older.

Pharmacology

Adapalene binds to specific retinoic acid nuclear receptors but does not bind to cytosolic receptor protein. Biochemical and pharmacological profile studies have demonstrated that Adapalene is a modulator of cellular differentiation, keratinization and inflammatory processes. Adapalene binds with specific retinoic acid nuclear receptors that normalize the differentiation of follicular epithelial cells resulting in decreased microcomedone formation. Benzoyl peroxide is an oxidizing agent with bacteriocidal and keratolytic effects.

Mechanism of action

Adapalene normalizes skin cell structure and helps unclog pores. Benzoyl peroxide kills bacteria and removes dead skin cells. Together, they target multiple causes of acne.

Dosage

Patients 12 years and older:

  • Apply a thin film of gel to affected areas of the face and/or trunk once daily at bedtime
  • Use a pea-sized amount for each area of the face (e.g., forehead, chin, each cheek)
  • Avoid eyes, lips and mucous membranes
Administration
  • For external use only (do not use in eyes, mouth, or vagina)
  • Clean and dry the affected area before application
  • Wash hands after applying (unless hands are the affected area)
  • Apply at bedtime
Missed dose

If you miss a dose, apply it as soon as you remember. If it is almost time for your next dose, skip the missed dose and continue with your regular schedule. Do not double the dose.

Side effects

Common side effects:

  • Erythema (skin redness)
  • Scaling (skin peeling)
  • Dry skin
  • Stinging/burning at application site
  • Skin irritation
  • Contact dermatitis
  • Application site burning
Precautions & warnings
  • Avoid exposure to sunlight and sunlamps (skin becomes more sensitive)
  • Use sunscreen and protective apparel (e.g., hat) when sun exposure cannot be avoided
  • Do not apply to broken, sunburnt or sensitive areas of skin
  • May bleach hair, clothing, and furnishings (be careful when applying to large areas)
  • Rinse with water immediately if contact with eyes or mouth occurs
  • Consult your doctor if pregnant, planning pregnancy or breastfeeding
Contraindications

Hypersensitivity to any component.

Drug interactions

Concomitant topical acne therapy should be used with caution because a possible cumulative irritancy effect may occur, especially with the use of peeling, desquamating, or abrasive agents. No formal drug-drug interaction studies were conducted.

Food interactions

No food interactions (topical application).

Use in pregnancy

Pregnancy Category: C. No adequate controlled studies in pregnant women. Use only if potential benefit justifies risk to the fetus.

Pregnancy Category: C — always consult a doctor before taking during pregnancy.

Use in lactation

It is not known whether Adapalene or Benzoyl peroxide is excreted in human milk. Use with caution.

Pediatric use

Under 12 years: Safety and efficacy have not been established.

12 years and older: Adult dose applies.

Geriatric use

No specific dose adjustment needed for elderly patients.

Renal / hepatic impairment

No dose adjustment needed for topical use (minimal systemic absorption).

Overdose effects

Topical overdose is unlikely; excessive application may cause skin irritation.

Storage conditions

Store below 25°C in a cool and dry place, protected from light and moisture. Keep out of reach of children. Keep the tube tightly closed after use.
