Description
Calcium Orotate is a calcium supplement containing calcium with a functional amino acid chelating ligand - orotic acid. Orotic acid assists the transport of calcium through cellular membrane structures, thus facilitating the intracellular uptake of calcium, particularly in bone. Calcium Orotate also helps in the maintenance of healthy cartilage.
Orotic acid is involved in the synthesis of DNA and RNA. Calcium Orotate gets high marks because of its ability to penetrate complex cell membranes so that it can be metabolized in cartilage. It acts against inflammatory diseases, helps in weight control, reduces the effects of mood swings, enhances cognitive function, protects the heart by enhancing cardiac muscle efficiency, and may minimize the risk of colon cancer.

Pharmacology
Calcium Orotate is used to prevent and treat calcium deficiency. Orotic acid assists the transport of calcium through cellular membrane structures, facilitating intracellular uptake of calcium, particularly in bone. Orotic acid is involved in DNA and RNA synthesis and helps maintain healthy cartilage.
Mechanism of action
The orotic acid component of Calcium Orotate penetrates cell membranes to help calcium reach inside cells, improving calcium absorption and utilization, especially in bone and cartilage.
Dosage
Calcium Orotate 400 mg: 2-3 tablets daily with meals or as directed by a physician.
Calcium Orotate 740 mg: 1-2 tablets daily with meals or as directed by a physician.
Note: Dosage is determined by the physician based on the patient's clinical condition.
Administration
Take with meals (for better absorption). Swallow tablets whole; do not chew or crush. Follow the dose and duration as advised by your physician.
Missed dose
If you miss a dose, take it as soon as you remember. Skip the missed dose if it is almost time for your next dose. Do not double the dose.
Side effects
Common side effects: Bloating and swelling in the abdomen.
Infrequent: Loss of appetite, upset stomach, constipation, nausea, vomiting, unusual weight loss, increased thirst/urination, weakness, unusual tiredness, formation of kidney stones.
Precautions & warnings
- Precaution is needed if the patient is allergic to calcium orotate.
- Caution in kidney disease, kidney stones, achlorhydria, disease of the pancreas, sarcoidosis, malabsorption syndrome, and heart disease.
- Caution during pregnancy and lactation (calcium requirements increase).

Drug interactions
- Bisphosphonates (alendronate), quinolone antibiotics (ciprofloxacin, levofloxacin), tetracycline antibiotics (doxycycline, minocycline): Calcium reduces absorption when taken together.
- Levothyroxine, phenytoin, tiludronate disodium: Absorption is reduced.
- Thiazide-type diuretics: Increased risk of hypercalcemia and hypercalciuria.
- Aluminum- and magnesium-containing antacids: Increase urinary calcium excretion.
- Mineral oil and stimulant laxatives: Decrease calcium absorption.
- Glucocorticoids (prednisone): Cause calcium depletion and osteoporosis with long-term use.
- Oral contraceptives and estrogen compounds: Reduce calcium.
- NSAIDs, aspirin, ibuprofen, corticosteroids: Deplete calcium.
Food interactions
Take with meals for better absorption; foods containing oxalic acid and phytic acid may reduce calcium absorption.
Use in pregnancy
Calcium requirements increase during pregnancy; calcium orotate may help reduce the risk of pregnancy-related high blood pressure. Consult your doctor.
Use in lactation
Calcium requirements increase during lactation; use under medical supervision.
Pediatric use
Use in children under medical supervision.
Geriatric use

Renal / hepatic impairment
Hepatic impairment: No special precautions required, but consult your doctor.
Overdose effects
Overdose may cause hypercalcemia, hypercalciuria, kidney stone formation. Symptoms: nausea, vomiting, constipation, increased thirst/urination, confusion. Consult your doctor immediately if overdose is suspected.
