Description
Dinoprostone is a naturally-occurring biomolecule that functions as a local hormone. It causes the cervix muscles to dilate and become relaxed and encourages uterine muscle contraction during labor. Its functions are similar to that of prostaglandin E2.

Pharmacology
Dinoprostone is a naturally-occurring biomolecule that functions as a local hormone. It causes the cervix muscles to dilate and become relaxed and encourages uterine muscle contraction during labor.
Mechanism of action
Dinoprostone relaxes cervical muscles and induces uterine contractions to initiate labor and induce abortion.
Dosage
Adult: 0.5 mg Dinoprostone gel (intracervical) as a single dose. Patient should remain in dorsal position for 10-15 minutes after placement.
Administration
Administered as an intracervical gel (by healthcare professional only).

This medicine is administered by healthcare professionals. You cannot miss a dose yourself.
Side effects
Common: Abnormal labor affecting fetus, fetal heart rate disorder, fetal distress syndrome, uterine hypertonus.
Uncommon: Nausea, vomiting, fever, abdominal pain, diarrhea.
Rare: Disseminated intravascular coagulation, uterine rupture, anaphylactic reaction.
Precautions & warnings
Caution in patients with hypertonus, sustained uterine contractility or fetal distress, asthma, glaucoma, heart diseases, depression.
Contraindications
Hypersensitivity; patients in whom oxytocic drugs are contraindicated; history of cesarean section or major uterine surgery; major cephalopelvic disproportion; history of difficult labor; grand multiparae (≥6 previous term pregnancies); pre-existing fetal distress; uterine overdistention; ruptured amniotic membranes; unexplained vaginal bleeding; fetal malpresentation; epilepsy; abnormal placenta or umbilical cord position.
Drug interactions
Oxytocic medications: Concurrent use is not advised (may increase action); allow at least 30 minutes interval before oxytocin use.
Alcohol: Do not consume alcohol while using Dinoprostone.
Food interactions
Dinoprostone interacts with alcohol; do not consume alcohol while using.
Use in pregnancy
Pregnancy Category C. Not recommended during pregnancy, labor, or breastfeeding.
Pregnancy Category: C — always consult a doctor before taking during pregnancy.
Use in lactation
Not recommended during breastfeeding.
Pediatric use
Not studied in pediatric population; not recommended.
Geriatric use
Not studied in patients over 65 years; not recommended.
Renal / hepatic impairment
No special precautions are necessary for patients with kidney or liver problems.
Overdose effects
Treatment of overdose is symptomatic.
