Description
Retapamulin is a pleuromutilin antibacterial isolated from the fungus Clitopilus passeckerianus. It is a bacteriostatic agent that inhibits bacterial protein synthesis by binding to protein L3 on the ribosomal 50S subunit. It inhibits peptidyl transfer and blocks P-site interactions, preventing the formation of active 50S ribosomal subunits.

Pharmacology
Retapamulin binds to protein L3 on the ribosomal 50S subunit, inhibiting bacterial protein synthesis. It inhibits peptidyl transfer and blocks P-site interactions, preventing the formation of active 50S ribosomal subunits.
Mechanism of action
Prevents bacterial growth by inhibiting bacterial protein synthesis.
Dosage
Adult Dose (Topical):
- 1% ointment: Apply to affected area twice daily for 5 days
- Re-evaluate if no response after 3 days of treatment
- Total treatment area: <100 cm² total body surface area
Pediatric Dose (Topical):
- 1% ointment: >9 months: Apply to affected area twice daily for 5 days
- Total treatment area: <2% of total body surface area
Administration
- For external use only
- Clean and dry the affected area before application
- Wash hands after applying (unless hands are the affected area)
- Do not use antimicrobial dressings
Missed dose
If you miss an application, apply it as soon as you remember; skip if it's almost time for next application; do not double the dose.

- Avoid contact with eyes, nose, mouth, or genital areas (rinse with water if contact occurs)
- Do not cover with airtight dressings (unless advised by doctor)
- Discontinue if infection worsens or redness/irritation increases
- Discard remaining medicine 7 days after opening
Contraindications
- Hypersensitivity to retapamulin or any ingredient
Drug interactions
- Concurrent application with other topical products to the same skin area is not recommended
