14 days daily administration for morning, mid-day and night dosing each containing Vonoprazan 20 mg and Amoxicillin 500 mg.

Pharmacology

Vonoprazan inhibits H+, K+-ATPase in gastric parietal cells, blocking gastric acid production. Amoxicillin is an antibacterial drug.

Mechanism of action

Vonoprazan blocks gastric acid production, and amoxicillin destroys H. pylori bacteria.

Dosage

Adult Dose (Oral):

  • Morning: 1 Vonoprazan 20 mg tablet & 2 Amoxicillin 500 mg capsules
  • Midday: 2 Amoxicillin 500 mg capsules
  • Night: 1 Vonoprazan 20 mg tablet & 2 Amoxicillin 500 mg capsules
  • With or without food, for 14 days
Administration
  • May be taken with or without food
  • Use as directed by a physician
Missed dose

If you miss a dose, take it as soon as you remember; if almost time for next dose, skip it; do not double the dose.

Side effects

Common Side Effects:

  • Diarrhea
  • Abdominal pain
  • Vulvovaginal candidiasis
  • Nasopharyngitis
Precautions & warnings
  • Serious and occasionally fatal hypersensitivity reactions (e.g., anaphylaxis) have been reported
  • Discontinue at the first signs or symptoms of SCAR
  • Evaluate if diarrhea occurs with Vonomax Duo
Contraindications
  • Known hypersensitivity to Vonoprazan, Amoxicillin or any other beta-lactams, or any component of Vonomax Duo
  • Rilpivirine-containing products
Drug interactions
  • Strong or moderate CYP3A inducers: drug-drug interaction may occur
  • Probenecid, Itraconazole, Allopurinol, Omeprazole, Saquinavir, Ritonavir, Etravirine & Rilpivirine
Food interactions

Food interactions are not significant.

Use in pregnancy

Use in Pregnancy: Based on findings from animal studies and observational studies in pregnant women with use of Clarithromycin, use of Vonomax Duo is not recommended in pregnant women.

Pregnancy Category: C — always consult a doctor before taking during pregnancy.

Use in lactation

Use in Lactation: Avoid breastfeeding during Vonomax Duo therapy.

Pediatric use

Safety and effectiveness not established in pediatric patients

Geriatric use

Caution required in elderly patients

Renal / hepatic impairment

Renal Impairment: Mild to moderate (eGFR 30-89 mL/min): no dose adjustment; severe (eGFR <30 mL/min): avoid

Hepatic Impairment: Mild: no dose adjustment; moderate to severe: avoid

Overdose effects

Symptoms: No information is available on accidental overdosage in humans.
