Zavegepant nasal spray is indicated for the acute treatment of migraine with or without aura in adults.
Description
Zavegepant is a calcitonin gene-related peptide (CGRP) receptor antagonist. The relationship between pharmacodynamic activity and the mechanism by which zavegepant exerts its clinical effects is unknown.

Pharmacology
Zavegepant is a calcitonin gene-related peptide (CGRP) receptor antagonist.
Mechanism of action
Antagonizes CGRP receptors, reducing migraine pain.
Dosage
Adult Dose (Nasal Spray):
- 10 mg given as a single spray in one nostril, as needed
- Maximum dose in a 24-hour period is 10 mg (one spray)
- Safety of treating more than 8 migraines in a 30-day period has not been established
Administration
- For nasal use only
- Avoid use of intranasal decongestants; if unavoidable, administer at least 1 hour after Zavegepant administration
Missed dose
If you miss a dose, take it as soon as you remember; if almost time for next dose, skip it; do not double the dose.

Precautions & warnings
- If a serious hypersensitivity reaction occurs, discontinue
- New-onset or worsening of pre-existing hypertension may occur
- New-onset or worsening of pre-existing Raynaud's phenomenon may occur
Contraindications
- History of hypersensitivity reaction to zavegepant or any of the components of this nasal spray
Drug interactions
- Avoid use with drugs that inhibit or induce OATP1B3 or NTCP transporters
- Intranasal decongestants: avoid use; if unavoidable, administer at least 1 hour after Zavegepant administration

Food interactions are not significant.
Use in pregnancy
Use in Pregnancy: There are no adequate data on the developmental risk associated with the use of Zavegepant in pregnant women.
Pregnancy Category: C — always consult a doctor before taking during pregnancy.
Use in lactation
Use in Lactation: There are no data on the presence of zavegepant or its metabolites in human milk.
