Azacitidine is a nucleoside metabolic inhibitor used for the treatment of myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML). It works by causing DNA hypomethylation and direct cytotoxicity on abnormal hematopoietic cells.
Indications
- Myelodysplastic Syndromes (MDS): Refractory anemia (RA), refractory anemia with ringed sideroblasts (RARS), refractory anemia with excess blasts (RAEB), refractory anemia with excess blasts in transformation (RAEB-T), and chronic myelomonocytic leukemia (CMMoL)
- Acute Myeloid Leukemia (AML) - maintenance therapy in adult patients who achieved complete remission after induction therapy
Dosage
Injection (MDS): 75 mg/m², subcutaneous or intravenous infusion, daily for 7 days, 28-day cycle; may increase to 100 mg/m² after 2 cycles if no benefit and no severe toxicity.
Oral Tablet (AML maintenance): 300 mg once daily, 14 days on treatment and 14 days off (28-day cycle).

Precautions & warnings
- Caution in hepatic or renal impairment
- May cause fetal harm during pregnancy

- Male patients should also use contraception during treatment
- Avoid use during breastfeeding
