Venetoclax is indicated in combination with Rituximab for the treatment of adult patients with chronic lymphocytic leukemia (CLL) who have received at least one prior therapy.
Indications
- Chronic lymphocytic leukemia (CLL)
Dosage
Adult Dose (Oral):
- Starting dose: 20 mg once daily for 7 days
- Gradually increased over 5 weeks up to 400 mg daily
- Post-titration dose in combination with Rituximab: 400 mg once daily; Rituximab started after 400 mg Venetoclax for 7 days
- Post-titration dose for monotherapy: 400 mg once daily; continue until disease progression or no longer tolerated
Side effects
Common Side Effects:
- Neutropenia
- Diarrhea
- Upper respiratory tract infection
- Nausea
- Anemia
- Fatigue
Precautions & warnings
- Tumour lysis syndrome (TLS) can occur; prophylaxis with hydration and anti-hyperuricemics needed
- Monitor blood chemistries
- Monitor CBC for neutropenia
- Do not administer live vaccines
- Avoid CYP3A inhibitors and inducers
