Lifitegrast eye prep is used for the treatment of the signs and symptoms of dry eye disease (DED). It binds to LFA-1 integrin on leukocytes, blocking interaction with ICAM-1 to reduce T-cell activation and inflammation.
Description
Lifitegrast binds to the integrin lymphocyte function-associated antigen-1 (LFA-1), a cell surface protein on leukocytes, and blocks the interaction of LFA-1 with its cognate ligand intercellular adhesion molecule-1 (ICAM-1). ICAM-1 may be overexpressed in corneal and conjunctival tissues in dry eye disease. LFA-1/ICAM-1 interaction can contribute to the formation of an immunological synapse resulting in T-cell activation and migration to target tissues. In vitro studies demonstrated that lifitegrast may inhibit T-cell adhesion to ICAM-1 and may inhibit secretion of inflammatory cytokines.

Pharmacology
Lifitegrast binds to LFA-1 integrin, blocking interaction with ICAM-1, which reduces T-cell activation and inflammatory cytokine secretion.
Mechanism of action
Relieves symptoms of dry eye disease by reducing T-cell-mediated inflammation.
Dosage
Adult Dose (Eye Drop):
- Instill 1 drop in each eye twice daily (approximately 12 hours apart)
Children (<17 years): Safety and efficacy not established
Administration
- For ophthalmic use only
- Drops are in single-use containers; discard immediately after using in each eye
- Remove contact lenses before instilling; may be reinserted 15 minutes after administration
- Do not touch the dropper tip to surfaces (to avoid contamination)
- Use within 1 month of opening
Missed dose
Not generally applicable (regular daily use); consult doctor if needed.
Side effects

- Instillation site irritation (5-25%)
- Dysgeusia (5-25%)
- Reduced visual acuity (5-25%)

Precautions & warnings
- Do not touch the dropper tip to surfaces (to avoid contamination)
- Remove contact lenses before instilling and reinsert 15 minutes after

- Limited data on use during pregnancy and lactation
