Description

Naloxone Hydrochloride is an opioid antagonist. It is used for the treatment of opioid toxicity. It helps to reverse the depressant effect on breathing, and on other parts of the body. It also helps to determine if a patient has taken the overdose of opioid. Naloxone Hydrochloride is given as an injection into veins, muscle or under the skin by a doctor or nurse. It is advisable not to self-administer the injection. Your doctor or nurse will train your family member or caregiver about how to inject the medicine in case of an emergency. It is advisable to follow your doctor's advice while being on the treatment. The most common side effects of this medicine include injection site redness, dizziness, headache and increased or decreased blood pressure. Most of these usually go away within a short time of the injection. If you are bothered by them or they go on for longer, talk to your doctor or nurse. You should not drive if this medicine makes you feel dizzy. Before taking it, tell your doctor if you have heart problems, high blood pressure or high heart rate. This may affect your treatment. Pregnant or breastfeeding women should consult their doctor before taking this medicine. Some other medicines you are taking may interfere with it, so make sure your doctor knows about all the other medicines you are using. Naloxone Hydrochloride is an opioid antagonist. Opioid medicines are strong painkillers, which in high doses can cause life-threatening side effects such as difficulty in breathing. Naloxone Hydrochloride works by blocking the effects of opioids leading to reversal of these side effects.

Mechanism of action

Naloxone is a pure opioid antagonist that acts competitively at opioid receptors.

Dosage

Adult Dose: Oral Opioid dependence Adult: As hydrochloride: 200 mg to 3 g daily. Intravenous Opioid overdosage Adult: As hydrochloride: 0.4-2 mg repeated if necessary at 2-3 min intervals. If there is no response after a total of 10 mg has been given, consider the possibility of overdosage with other drugs. Reduce dose for opioid-dependent patients: 0.1-0.2 mg. IM/SC routes may be used (at IV doses) if IV admin is not feasible. Reversal of central depression from opioid use during surgery Adult: As hydrochloride: 100-200 mcg at intervals of 2-3 minute, titrate dose according to response while maintaining analgesia. Child Dose: Intravenous Opioid overdosage Child: As hydrochloride: Initially 10 mcg/kg IV followed by 100 mcg/kg IV if necessary. Alternatively, 0.4-0.8 mg IM or SC, repeated as necessary, if IV admin is not feasible. Parenteral Opioid-induced depression in neonates due to obstetric analgesia Child: As hydrochloride: 10 mcg/kg IV, IM or SC repeated at 2-3 min intervals if necessary or 60 mcg/kg as a single IM dose. Intravenous Reversal of central depression from opioid use during surgery Child: As hydrochloride: 5-10 mcg IV at 2-3 min intervals.

Precautions & warnings

[SAFE] Naloxone Hydrochloride does not usually affect your ability to drive. It is given as an injection into veins, muscle or under the skin by the healthcare professionals only. Naloxone Hydrochloride might make you feel dizzy. If this happens, avoid driving or operating on machinery. Inform the doctor if you are taking any pain killers, sleeping pills (sedatives), narcotic painkillers, or other medicines Avoid consuming alcohol while taking the medicine as it can affect the working of the medicine.

Hypersensitivity.

Drug interactions

Decreased effect of opioid analgesics. Potentially Fatal: Acute withdrawal reaction in physically dependent patients may occur with opioid analgesics. Severe cardiopulmonary effects may occur with cardiotoxic drugs.

Food interactions

[UNSAFE] It is unsafe to consume alcohol with Naloxone Hydrochloride.

Use in pregnancy

[SAFE IF PRESCRIBED] Naloxone Hydrochloride is generally considered safe to use during pregnancy. Animal studies have shown low or no adverse effects to the developing baby; however, there are limited human studies.

Use in lactation

[SAFE IF PRESCRIBED] Naloxone Hydrochloride is probably safe to use during breastfeeding. Limited human data suggests that the drug does not represent any significant risk to the baby.

Renal / hepatic impairment

[SAFE IF PRESCRIBED] Naloxone Hydrochloride is probably safe to use in patients with kidney disease. Limited data available suggests that dose adjustment of Naloxone Hydrochloride may not be needed in these patients. Please consult your doctor.
