Pharmacology
Allystrenol is a synthetic progestin that acts like progesterone. It improves embryo attachment to the uterine lining and enhances blood flow to the placenta. It relaxes uterine muscles to prevent premature labor. It also helps in placental hormone production (hCG, hPL, estrogen).
Mechanism of action
Allystrenol acts like progesterone. It prepares the uterine lining, helps maintain pregnancy by reducing uterine sensitivity to oxytocin and preventing premature labor.
Dosage
Adults:
- IUGR: 5 mg 3 times daily for at least 2 months (reduce dose if symptoms improve)
- Threatened abortion: 5 mg 3 times daily until symptoms disappear
- Habitual abortion: 5-10 mg/day (1-2 tablets) as soon as pregnancy is diagnosed, continue for at least 1 month after the critical period
- Threatened premature delivery: Up to 40 mg daily as per physician's advice
Administration
May be taken with or without food (take with meals if GI upset occurs). Swallow tablet whole, do not crush. Take at a fixed time each day.
Missed dose
If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and continue with your regular schedule. Do not double the dose.
Side effects
Common side effects: Edema (swelling), bloating, anxiety, irritation, depression, muscle pain, nausea, vomiting, skin rash, headache, breast changes
Precautions & warnings
- Discontinue after delivery (may affect the baby)
- Do not stop suddenly without consulting your doctor
- Caution while driving (may cause dizziness)
- Caution in diabetes, epilepsy, asthma, cardiac or renal dysfunction

Drug interactions
- Enzyme-inducing drugs (carbamazepine, griseofulvin, phenobarbital, phenytoin, rifampicin): Increase clearance
- Ketoconazole: May increase bioavailability
- Cyclosporine: May increase concentration (potentially fatal)
