Amoxicillin + Clarithromycin + Lansoprazole Kit

Amoxicillin + Clarithromycin + Lansoprazole Kit is a combination medicine used in the treatment of peptic ulcer disease. It contains two antibiotics (Amoxicillin and Clarithromycin) and a proton pump inhibitor (Lansoprazole).

Amoxicillin inhibits bacterial cell wall synthesis, Clarithromycin inhibits bacterial protein synthesis, and Lansoprazole reduces gastric acid secretion to help ulcer healing. Together, they effectively eliminate H. pylori infection.

This medicine should be taken on an empty stomach. Complete the full course as prescribed - do not stop even if you feel better. Common side effects include nausea, diarrhea, stomach pain, flatulence, metallic taste, dry mouth, headache, and skin rash.

Lansoprazole: Proton pump inhibitor that binds to H+/K+-ATPase (proton pump) in gastric parietal cells, blocking acid secretion. Amoxicillin: Inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins, inhibiting the final transpeptidation step of peptidoglycan synthesis, resulting in bacterial lysis. Clarithromycin: Inhibits protein synthesis by binding to the 50S ribosomal subunit, causing antibacterial activity.

Amoxicillin destroys bacteria by inhibiting cell wall formation. Clarithromycin prevents bacterial protein synthesis, stopping bacterial growth. Lansoprazole reduces stomach acid secretion, helping ulcers heal. Together, these three medicines effectively eliminate H. pylori infection.

Adult Dosage:

• Amoxicillin: 500 mg + Clarithromycin: 250 mg + Lansoprazole: 30 mg
• Twice daily (every 12 hours) for 10-14 days
• Dosage and duration as per physician's advice

Special Precautions:

• Contraindicated in severe penicillin hypersensitivity
• Clarithromycin contraindicated in QT prolongation or ventricular arrhythmia history
• Clarithromycin contraindicated in cholestatic jaundice/hepatic dysfunction history
• Not recommended in patients with CrCl <30 mL/min

Renal Dose: Dose adjustment may be needed in severe renal impairment.

Take this medicine on an empty stomach as advised by your doctor. Follow the prescribed dosage and duration. Complete the full course of treatment.

If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and continue with your regular schedule. Do not double the dose.

Important Precautions:

• Avoid driving or activities requiring mental focus
• Avoid alcohol consumption (may cause Disulfiram reaction)
• Consult doctor during pregnancy

• Severe hypersensitivity to any component
• History of cholestatic jaundice/hepatic dysfunction with clarithromycin use
• History of QT prolongation or ventricular cardiac arrhythmia (including torsades de pointes)
• Severe hypersensitivity reactions (e.g., anaphylaxis or Stevens-Johnson syndrome) to amoxicillin or other beta-lactam antibiotics
• Known hypersensitivity to clarithromycin, erythromycin, or any macrolide antibiotics
• CrCl less than 30 mL/min

• Allopurinol: Increases risk of skin rash
• Warfarin: May increase INR
• Simvastatin/Lovastatin: Clarithromycin increases their levels
• Methotrexate: May increase methotrexate levels
• Carbamazepine, Theophylline: Clarithromycin increases their levels
• Antacids: Maintain 1-hour gap with Lansoprazole

[UNSAFE] Consuming alcohol with Amoxicillin + Clarithromycin + Lansoprazole Kit may cause flushing, increased heart beat, nausea, thirst, chest pain and low blood pressure (Disulfiram reaction).

[CONSULT YOUR DOCTOR] Amoxicillin + Clarithromycin + Lansoprazole Kit may be unsafe to use during pregnancy. Animal studies have shown harmful effects on the developing baby. Your doctor will weigh the benefits and risks before prescribing.

[SAFE IF PRESCRIBED] Amoxicillin + Clarithromycin + Lansoprazole Kit is probably safe to use during breastfeeding. Limited human data suggests that the drug does not represent any significant risk to the baby.

Safety and effectiveness in pediatric patients have not been established.

Elderly patients may have decreased renal function, so dose adjustment may be needed.

[CAUTION] Use with caution in kidney disease. Not recommended in severe kidney disease (CrCl <30 mL/min).

[SAFE IF PRESCRIBED] Probably safe in liver disease. Dose adjustment may not be needed, but consult your doctor.

Overdose may cause nausea, vomiting, diarrhea, and electrolyte imbalance. Consult your doctor if symptoms occur.
