Pharmacology

Glucosamine stimulates the production of proteoglycans and increases sulfate uptake by articular cartilage. Diacerein is an anthraquinone derivative that acts via inhibition of interleukin-1 in osteoarthritis.

Mechanism of action

Glucosamine supports cartilage formation; Diacerein inhibits interleukin-1 to reduce inflammation and cartilage destruction.

Dosage

Adults and Children (>12 years): 1 tablet twice daily with food.
Renal impairment (CrCl <30): 25 mg daily.

Administration

Take with food or immediately after meals.

Missed dose

If you miss a dose, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose.

Side effects

Common: Diarrhea, constipation, epigastric pain, heartburn, nausea, vomiting, headache, skin rashes, drowsiness, intense yellow coloring of urine.

Precautions & warnings

Caution in patients allergic to glucosamine, diacerein or shellfish; those with diabetes, asthma, alcohol dependence or liver disease. Regular monitoring of blood count, liver function and urinalysis is recommended.

Contraindications

Hypersensitivity; pregnancy and lactation.

Drug interactions

Aluminium/magnesium hydroxide antacids: Decreased absorption of diacerein.
Laxatives, antibiotics: Increased risk of diarrhea.
Fibers and phytates: Avoid co-administration.
Antidiabetic agents, doxorubicin, etoposide: May reduce effect.
Warfarin: May enhance anticoagulant activity.
