Sibutramine Hydrochloride is an anti-obesity medication used for weight management in obesity. It inhibits reuptake of norepinephrine, serotonin, and dopamine to increase satiety and reduce appetite.
Description
Sibutramine produces its therapeutic effects by inhibiting reuptake of norepinephrine (NE), serotonin (5-HT), and to a lesser extent, dopamine at the neuronal synapse. By inhibiting reuptake of these neurotransmitters, it promotes satiety and decreases appetite, leading to reduced food intake and weight loss.

Pharmacology
Sibutramine inhibits reuptake of norepinephrine, serotonin, and dopamine at the neuronal synapse. Increased levels of these neurotransmitters promote satiety, reduce appetite, and decrease food intake, leading to weight loss.
Mechanism of action
Reduces appetite and increases satiety by inhibiting serotonin and norepinephrine reuptake.
Dosage
Adult Dose (Oral):
- Starting: 10 mg once daily (with or without food)
- Dose titration: Increase to 15 mg/day after 4 weeks if inadequate weight loss
- 5 mg dose: For patients who do not tolerate the 10 mg dose
- Maximum: 15 mg/day; doses above 15 mg/day are not recommended
Administration
- Take orally with or without food
- Take at the same time each day
- Take as directed by a physician
Missed dose
If you miss a dose, take it as soon as you remember; if almost time for the next dose, skip it; do not double the dose.

- Constipation

Precautions & warnings
- Do not use in history of coronary artery disease, CHF, tachycardia, peripheral arterial disease, arrhythmia, or cerebrovascular disease
- Do not use in uncontrolled hypertension >145/90 mmHg
- Do not use in patients over 65 years
- Do not use with MAOIs
- Do not use in major eating disorders (anorexia nervosa or bulimia)
Contraindications
- History of coronary artery disease, CHF, tachycardia, peripheral arterial disease, arrhythmia, or cerebrovascular disease
- Uncontrolled hypertension >145/90 mmHg
- Age over 65 years
- Receiving MAOIs
- Hypersensitivity to sibutramine or any component
- Major eating disorders
- Taking other centrally acting weight loss drugs
Drug interactions
Possible interactions with other serotonergic drugs (risk of serotonin syndrome).
Food interactions
Caution is advised when consuming alcohol with Sibutramine; consult your doctor.
