Tioconazole Vaginal Prep is an antifungal used for vulvovaginal candidiasis (vaginal fungal infection). It kills fungi by destroying their cell membrane.
Description
Tioconazole is a synthetic azole antifungal that interferes with ergosterol synthesis, needed for fungal cell membrane synthesis. It exhibits fungicidal activity in vitro against Candida albicans, other Candida species, and Torulopsis glabrata. It is effective as a single-dose treatment for vulvovaginal candidiasis.

Pharmacology
Tioconazole inhibits ergosterol synthesis, disrupting fungal cell membrane integrity. Ergosterol is an essential component of fungal cell membranes; its deficiency increases membrane permeability and causes fungal cell death.
Mechanism of action
Destroys fungal cell membranes by inhibiting ergosterol synthesis.
Dosage
Adult Dose (Vaginal):
- Vulvovaginal candidiasis: One full applicator (about 4.6 g) intravaginally at bedtime as a single dose
- Or 300 mg suppository intravaginally as a single dose
Pediatric Dose (>12 years):
- Same as adult dose
Administration
- For intravaginal use only
- Administration at bedtime is preferred
- Wash hands before application
- Use applicator to administer intravaginally
Missed dose
If you miss a dose, apply at the next scheduled time.
Side effects
Common Side Effects:
- Vaginal burning
- Itching
- Irritation
- Discharge, vulvar edema and swelling
- Vaginal pain, dysuria, nocturia, dyspareunia
- Dryness of vaginal secretions, desquamation
Precautions & warnings
- For intravaginal administration only
- Can damage condoms and diaphragms; do not use these within 72 hours of treatment
- Avoid contact with eyes
- Inform doctor if no improvement after 3 days or if it lasts more than 7 days
Contraindications
- Hypersensitivity to imidazole-derivative azole antifungals
- Self-medication in women with diabetes or HIV infection
- Girls under 12 years
Drug interactions
- Cyclosporine: Hypersensitivity

Alcohol consumption is not relevant (topical application).
