Netarsudil + Latanoprost is an eye drop used for reducing elevated intraocular pressure (IOP) in open-angle glaucoma or ocular hypertension. Netarsudil is a Rho kinase inhibitor and Latanoprost is a Prostaglandin F2 analogue.
Description
This is a fixed-dose combination of a Rho kinase inhibitor and a Prostaglandin F2 analogue. Netarsudil works at the cellular level within the trabecular outflow pathway to relax actin-myosin, causing relaxation within the trabecular meshwork and inner wall of Schlemm's canal, and decreases episcleral venous pressure (EVP). Latanoprost is a selective FP receptor agonist which reduces IOP by increasing uveoscleral outflow.

Pharmacology
Netarsudil inhibits Rho kinase to relax trabecular meshwork and inner wall of Schlemm's canal, decreasing episcleral venous pressure. Latanoprost acts as FP receptor agonist to increase uveoscleral outflow and reduce IOP.
Mechanism of action
Reduces intraocular pressure by increasing trabecular and uveoscleral outflow.
Dosage
Adult Dose (Eye Drops):
- 1 drop in affected eye(s) once daily in the evening
Administration
- For ophthalmic use only
- Do not touch dropper tip to eye or any surface
- Remove contact lenses before administration (reinsert after 15 minutes)
- Administer at least 5 minutes apart from other eye drops
Missed dose
If you miss a dose, use it as soon as you remember; skip if it's almost time for next dose; do not double the dose.
Side effects
Common Side Effects:
- Conjunctival hyperemia (53%)
- Corneal verticillata (~20%)
- Instillation site pain
- Conjunctival hemorrhage
- Instillation site erythema (5-10%)
- Corneal staining
- Blurred vision
- Increased lacrimation
- Erythema of eyelid
- Reduced visual acuity
Precautions & warnings
- Caution in active intraocular inflammation (iritis/uveitis)
- May cause changes to pigmented tissues
- May gradually change eyelashes and vellus hair in treated eye
- Cystoid macular edema may occur
- Reactivation of Herpes Simplex keratitis may occur
Contraindications
- Hypersensitivity to any component
Drug interactions
Administer at least 5 minutes apart from other topical ophthalmic products.
Food interactions
No food interactions (eye drops).
Use in pregnancy
Use in Pregnancy: No adequate controlled studies in pregnant women; systemic exposure from ocular administration is low.
Pregnancy Category: C — always consult a doctor before taking during pregnancy.
Use in lactation
Use in Lactation: No data on presence in human milk; use with caution.
Pediatric use
Safety and effectiveness not established in pediatric patients.
Geriatric use
No overall differences in safety and effectiveness between elderly and other adult patients.
Renal / hepatic impairment
Renal Impairment: Caution in kidney disease; dose adjustment may be needed.

Storage conditions
Before opening: Store at 2-8°C in a dry place, protect from light. After opening: May be kept at 2-25°C for up to 6 weeks.
