Ofloxacin 0.3% E/E drops is an antibiotic used for bacterial eye infections, corneal ulcer, chronic suppurative otitis media and otitis externa. It inhibits bacterial DNA gyrase enzyme to prevent bacterial division and repair.
Description
Ofloxacin is a fluoroquinolone antibiotic that inhibits bacterial topoisomerase IV and DNA gyrase enzymes required for DNA replication, transcription, repair and recombination. It has activity against a wide range of gram-negative and gram-positive microorganisms.

Pharmacology
Ofloxacin inhibits bacterial topoisomerase IV and DNA gyrase enzymes to prevent DNA replication, transcription, repair and recombination, resulting in bacterial cell death.
Mechanism of action
Treats eye and ear infections by inhibiting bacterial DNA replication.
Dosage
Eye Drops (Adult):
- Bacterial conjunctivitis: 1-2 drops every 2-4 hours (first 2 days), then 4 times daily; max 10 days
- Corneal ulcer: Days 1-2: 1-2 drops q30min while awake; Days 3-7: 1-2 drops q1hr; Days 7-9: 4 times daily
Ear Drops:
- Chronic suppurative otitis media (12 years+): 10 drops twice daily for 14 days
- Otitis externa: 10 drops once daily for 7 days; (6 months-13 years: 5 drops once daily)
Administration
- For external use only
- Do not touch dropper tip to eye/ear
- Instill in lower eyelid
- Ear: Tilt head, keep ear facing up for 5 minutes
Missed dose
If you miss a dose, use it as soon as you remember; skip if it's almost time for next dose; do not double the dose.
Side effects
Common Side Effects:
- Eye discomfort
- Eye irritation
- Eye pain
- Blurred vision
Precautions & warnings
- Do not wear contact lenses until infection clears
- Apply pressure on corner of eye (near nose) for 1 minute after instillation
- Use within 4 weeks of opening
- Caution while driving (may cause blurred vision)
Contraindications
- Hypersensitivity to ofloxacin, other quinolones, or any component
Drug interactions
- Risk of QT prolongation with Class IA/III antiarrhythmics
- Decreased serum/urine concentrations with Mg/Al/Ca-containing antacids
- Increased risk of tendon disorders with corticosteroids
- Increased risk of CNS stimulation (seizures) with NSAIDs
Food interactions
No food interactions (eye/ear drops).
Use in pregnancy
Use in Pregnancy: Ofloxacin eye/ear drops may be unsafe during pregnancy; animal studies show evidence of fetal harm.
Pregnancy Category: C — always consult a doctor before taking during pregnancy.
Use in lactation
Use in Lactation: Ofloxacin eye/ear drops are probably unsafe during breastfeeding; may pass into breast milk and harm the baby.
Pediatric use
Eye drops: 1 year+; Ear drops: Otitis externa 6 months+, Chronic suppurative otitis media 12 years+.
Geriatric use
No dose adjustment required in elderly.
Renal / hepatic impairment
Renal Impairment: Caution in kidney disease; dose adjustment may be needed.
Hepatic Impairment: Caution in liver disease; dose adjustment may be needed.
Overdose effects
Overdose unlikely (topical use); symptomatic treatment.

Store at room temperature; protect from light; keep out of reach of children.
