Description
Pyridoxine is a water-soluble vitamin that functions in the metabolism of carbohydrates, proteins, and fats. It is essential for Hb formation and GABA synthesis within the CNS. It also aids in the release of glycogen stored in the liver and muscles. Vitamin B6 affects cellular and humoral responses of the immune system and plays a role in lymphocyte differentiation and maturation, antibody production, and lymphocyte proliferation.

Pharmacology
Pyridoxine is a water-soluble vitamin that functions as a coenzyme in the metabolism of carbohydrates, proteins, and fats. It is essential for Hb formation and GABA synthesis within the CNS. It plays a role in immune system function, lymphocyte differentiation and maturation, antibody production, and lymphocyte proliferation.
Mechanism of action
Functions as a coenzyme in carbohydrate, protein, and fat metabolism to correct vitamin B6 deficiency.
Dosage
Adult Dose (Oral):
- General deficiency: Up to 150 mg/day
- Sideroblastic anemia: Up to 400 mg/day
- Premenstrual syndrome: 50-100 mg/day
- Peripheral neuritis prophylaxis: 10-50 mg/day
- Peripheral neuritis treatment: 100-200 mg/day
- Nausea in pregnancy: 75 mg/day
Administration
- Take with food (preferably)
- Take at the same time each day
Missed dose
If you miss a dose, take it as soon as you remember; skip if it's almost time for next dose; do not double the dose.
Side effects
Side Effects:
- Severe peripheral neuropathy (with long-term large doses, excess of about 2g daily)
- Unstable gait
- Paresthesias
- Decreased sensation
- Decreased folic acid (with excessive doses)
Precautions & warnings
- Reduces the effects of levodopa; this does not occur if a dopa decarboxylase inhibitor is also given
- Isoniazid, penicillamine, and oral contraceptives may increase the requirement for pyridoxine
- Do not use in patients receiving levodopa
Contraindications
- Hypersensitivity to pyridoxine or any ingredient
- Patients receiving levodopa
Drug interactions
- Levodopa: Reduced therapeutic effect
- Phenobarbitone, phenytoin: Increased hepatic metabolism (40-50%), decreased serum concentration
- Isoniazid, penicillamine, oral contraceptives: Increased requirement for pyridoxine
Food interactions
It is not known whether it is safe to consume alcohol with Pyridoxine Hydrochloride.
Use in pregnancy
Use in Pregnancy: Safe during pregnancy; requirement increases during pregnancy; used for nausea and vomiting of pregnancy.
Pregnancy Category: A — always consult a doctor before taking during pregnancy.
Use in lactation
Use in Lactation: Safe during breastfeeding; requirement increases during lactation; caution required.
Pediatric use
Pediatric dose based on condition; consult your doctor.
Geriatric use
Caution required in elderly patients; dose adjustment may be needed.
Renal / hepatic impairment
Renal Impairment: Safe in kidney disease; no dose adjustment needed; consult your doctor.
Hepatic Impairment: Safe in liver disease; no dose adjustment needed; consult your doctor.
Overdose effects
Overdose: Severe peripheral neuropathy; treatment: discontinue drug, symptomatic supportive care.
