Silodosin is an alpha-1 adrenergic receptor antagonist used for the treatment of signs and symptoms of benign prostatic hyperplasia (BPH). It relaxes muscles in the prostate and bladder to improve urine flow.

Pharmacology
Silodosin blocks post-synaptic alpha-1 adrenoreceptors in the prostate, bladder base, bladder neck, prostatic capsule, and urethra. This blockade relaxes smooth muscle, reduces urethral resistance, and improves urine flow. It improves BPH symptoms such as difficulty starting urination, weak stream, and frequent urination.
Dosage
Adult Dose (Oral):
- Recommended: 8 mg once daily with a meal
- Moderate renal impairment (CrCl 30-50): 4 mg once daily
Side effects
Common Side Effects:
- Retrograde ejaculation
- Orthostatic hypotension
- Dizziness, headache
- Nasal congestion
- Diarrhea
Use in pregnancy
Use in Pregnancy: Silodosin is not indicated for women; generally safe; animal studies show low or no adverse effects; consult your doctor.
Pregnancy Category: C — always consult a doctor before taking during pregnancy.
Precautions & warnings
- Postural hypotension (dizziness) may develop when starting treatment
- Do not use with other alpha-blockers
- Inform ophthalmologist before cataract surgery (IFIS risk)
- Do not use in severe renal or hepatic impairment
Overdose effects
Overdose may cause orthostatic hypotension; maintain supine position; consider IV fluids or vasopressors; dialysis is not effective (97% protein bound).
