Pharmacology
Inhibits viral helicase-primase complex essential for viral DNA replication, distinct from DNA polymerase inhibitors.
Dosage
Herpes simplex: 200 mg once daily after meals. Recurrent herpes simplex: 1200 mg single dose within 6 hours. Herpes zoster: 400 mg once daily after meals.
Side effects
Headache, nausea, diarrhea, abdominal pain, thrombocytopenia, gingival bleeding, palpitations.
Use in pregnancy
Animal studies showed no adverse fetal effects, but human data is limited.

Special monitoring essential for severe renal impairment, thrombocytopenia, and elderly patients.
