Dicycloverine Hydrochloride is an antispasmodic and anticholinergic drug used for the treatment of gastrointestinal tract spasm and irritable bowel syndrome (IBS).
Description
Dicycloverine HCl relieves smooth muscle spasm in the GI and urinary tract. This effect is partly due to antimuscarinic action and partly direct action on the smooth muscle. It works at specific cholinergic (muscarinic) receptors located on the involuntary muscle in the walls of the gut, causing gut muscle relaxation and relieving colic pain.

Pharmacology
Dicycloverine HCl relieves smooth muscle spasm in the GI and urinary tract. This effect is partly due to antimuscarinic action and partly direct action on the smooth muscle. It binds to cholinergic (muscarinic) receptors on the involuntary muscle in the gut wall, causing muscle relaxation.
Mechanism of action
Dicycloverine blocks muscarinic receptors and acts directly on smooth muscle to relax intestinal muscles and relieve spasm.
Dosage
Adult (Oral): 10-20 mg three times daily.
Adult (IM Injection): 20 mg 4 times daily (only for 1-2 days; switch to oral therapy as soon as possible).
Child (2-12 years): 10 mg three times daily.
Child (6 months-2 years): 5-10 mg 3-4 times daily.
Administration
May be taken with or without food.
Missed dose
If you miss a dose of Dicycloverine, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose. Do not double the dose.
Side effects
Common: Difficulty in accommodation, exacerbation of glaucoma, tachycardia, palpitations, arrhythmias, urinary retention, restlessness, confusion, excitement, hallucination and delirium.
Potentially Fatal: Respiratory arrest in infants <10 weeks.
Precautions & warnings
Caution in hepatic or renal disease, urinary retention, paralytic ileus, hyperthyroidism, hypertension, congestive heart failure, cardiac tachyarrhythmia, children, elderly, pregnancy.
Contraindications
Intestinal obstruction; intestinal atony; myasthenia gravis; glaucoma; reflux oesophagitis; infants <6 months; lactation.
Drug interactions
Metoclopramide: Antagonizes the effect of drugs affecting GI motility.
Antacids: Absorption may be affected.
Food interactions
Can be taken with or without food.
Use in pregnancy
Pregnancy Category B. Should be used during pregnancy only if clearly needed.
Pregnancy Category: B — always consult a doctor before taking during pregnancy.
Use in lactation
It is not known whether Dicycloverine is excreted in breast milk; use with caution.
Pediatric use
Contraindicated in infants under 6 months of age.
Geriatric use
Use with caution in elderly patients.
Renal / hepatic impairment
Caution in hepatic or renal disease.
Overdose effects
Overdose symptoms: headache, nausea, vomiting, blurred vision, dilated pupils, hot dry skin, dizziness, dry mouth, difficulty swallowing, CNS stimulation.
Storage conditions
Keep below 30°C temperature, away from light & moisture. Keep out of reach of children.
