Description
This ear drop contains antibacterial (ofloxacin), antifungal (clotrimazole), anti-inflammatory (beclomethasone) and local anesthetic (lidocaine). Ofloxacin inhibits DNA gyrase to prevent bacterial DNA replication. Clotrimazole inhibits ergosterol synthesis to reduce fungal cell wall integrity. Beclomethasone induces phospholipase A2 to inhibit arachidonic acid release, reducing inflammation. Lidocaine provides local anesthesia to reduce pain and stinging.

Pharmacology
Ofloxacin inhibits DNA gyrase to prevent bacterial DNA replication. Clotrimazole inhibits cytochrome P-450-mediated ergosterol synthesis to reduce fungal cell wall integrity. Beclomethasone induces phospholipase A2 to inhibit prostaglandin and leukotriene release. Lidocaine blocks sodium channels to provide local anesthesia.
Mechanism of action
Inhibits bacterial DNA replication and fungal cell wall formation, reduces inflammation and relieves pain.
Dosage
Adult and Pediatric Dose (Ear Drops):
- 2-5 drops 3-4 times daily for 7-14 days
- Otitis externa (intact tympanic membrane): 6 months+
- Chronic suppurative otitis media (perforated tympanic membrane): 12 years+
Administration
- Tilt head to one side so ear faces up
- Adults and >3 years: Pull ear lobe up and backward
- Children: Pull ear lobe down and backward
- Do not touch dropper tip to ear
Missed dose
If you miss a dose, use it as soon as you remember; skip if it's almost time for next dose; do not double the dose.

Precautions & warnings
- Discontinue and obtain cultures if no response in 1 week
- Prolonged use may result in overgrowth of nonsusceptible organisms
- Discontinue if local irritation or sensitization occurs
- If otorrhea persists or recurs, evaluate for cholesteatoma, foreign body, or tumor
- Not for ophthalmic use
- Not for injection

- Hypersensitivity to ofloxacin or other quinolones, or any component
- Viral infections of the ear

Overdose effects
